Raja Babu was born as Appala Raju to Punyamurthula Umamaheswara Rao and Ramanamma in Rajahmundry, East Godavari District, Andhra Pradesh. Chittibabu and Ananth Babu are his two brothers who are also well-known comedians. He completed his intermediate (10+2) schooling and the teacher training course. After that, he worked as a Telugu teacher for a while. He appeared in Dramas such as Kukka Pilla Dorikindi, Naalugilla Chaavadi, and Alluri Sitarama Raju during that time.

Garikapati Raja Rao contacted Babu after observing his acting talent in Mr. Gayudu’sproductioms. Babu arrived in Madras in 1960 and initially supported himself through tuition fees. In 1960, film director Addala Narayan Rao offered him a role in his film “Samajam”. Babu was well-known for his roles in slapstick comedies. In less than 20 years, he appeared in a total of 589 movies.
He performed as a hero in Tata Manavadu, PichodiPelli, Thirupathi, EvarikiVare Yamuna Teere, and Manishi Rodduna Paddadu, etc.
